ABSTRACT

Cardiovascular disease, such as coronary heart disease and acute myocardial infarction, is a leading cause of death globally. Due to the limited proliferative and regenerative capacity of adult mammalian cardiomyocytes (CMs), any of the current therapies cannot reverse the massive loss of CMs and subsequent fibrosis resulting from cardiac injury. Mammals mainly rely on glycolysis in the embryonic stage and fatty acid oxidation after birth for energy production. Recent reports have indicated that this metabolic pattern switch is closely related to the loss of CM proliferation. In this review, we summarize the biological characteristics of CMs and advances in heart regeneration, meanwhile shed light on the important role of CMs energy metabolism in cardiac regeneration.

ABSTRACT

AIM: To investigate the effect of lncRNA MALAT1 on the proliferation, migration and angiogenesis of retinal vascular endothelial cells and its molecular mechanism.METHODS: The expression levels of lncRNA MALAT1 in plasma of normal control group, diabetic without retinopathy group and diabetic retinopathy group were detected by qPCR and the effect of glucose culture on the expression levels of lncRNA MALAT1 were detected by qPCR too. The expression level of miR-124-3p was detected by qRT-PCR; Western blotting was used to detect the expression level of SOX7; The targeting relationship between lncRNA MALAT1 and miR-124-3p, miR-124-3p and SOX7 were detected by the dual-luciferase reporter system; CCK-8 assay was used to detect cell proliferation activity; Transwell assay was used to detect the migration ability of cells; Angiogenesis of hRMECs cells was measured by in vitro tube formation assay.RESULTS:The expression level of lncRNA MALAT1 in plasma of diabetic retinopathy patients was significantly higher than that of diabetic without retinopathy group and normal control group(P<0.001). In vitro glucose culture significantly promoted the expression of lncRNA MALAT1 in hRMECs cells, as well as the proliferation, migration and angiogenesis of hRMECs cells(all P<0.05). Knockdown of lncRNA MALAT1 significantly inhibited the proliferation, migration and tubule formation of hRMECs cells(all P<0.05). Dual-luciferase reporter gene assay showed that lncRNA MALAT1 targeted with miR-124-3p, and miR-124-3p targeted with SOX7. Overexpression of miR-124-3p significantly inhibited the proliferation, migration and tubule formation of hRMECs cells(all P<0.05). Overexpression of lncRNA MALAT1+miR-124-3p, miR-124-3p+SOX7, and knockdown of lncRNA MALAT1+overexpression of SOX7 all significantly eliminated the inhibitory effect of hRMECs cells(all P<0.05).CONCLUSION: lncRNA MALAT1 promote the proliferation, migration and angiogenesis of retinal endothelial cells in diabetic retinopathy by down-regulating the negative regulation of miR-124-3p on SOX7. Therefore, abnormal upregulation of lncRNA MALAT1 in patients with diabetic retinopathy is a potential biomarker.

ABSTRACT

@#Objective To identify the effectiveness of physical therapy on neuromuscular control dysfunction of chronic ankle instability (CAI). Methods CAI physiotherapy-related systematic reviews, meta-analysis, randomized controlled studies and case-controlled studies were systematically searched in PubMed, Web of Science, Cochrane, PEDro, EBSCO and CNKI from October 1st, 2010 to October 1st, 2020. After quality assessment, the research evidence regarding certain intervention was then graded according to the Oxford Center for Evidence-based Medicine. Results A total of 20 articles were included. Intervention based on unstable plane exercise training could significantly improve lower limb balance control ability (Ⅰ A) and muscle strength (Ⅱ A) for CAI patients. The efficiency of exercise-based intervention could be further raised (Ⅱ C) with the aid of external equipment, but either the benefits in muscle reaction (Ⅲ C) or proprioception (Ⅱ D) was limited. Six sessions or more of manual therapy could be used to enhance the balance ability of the lower limbs (Ⅱ B), while taping failed to provide improvements in ankle proprioception in short period or immediately (Ⅰ A). Conclusion Unstable plane training-based therapeutic exercise and manual therapy could effectively enhance the neuromuscular control with high-quality evidence supporting, which can be primary interventions for CAI patients. Meanwhile, taping and other external devices could be used as adjunct methods to improve the efficiency of physiotherapy program, while there is currently no evidence to support the application of modality treatment in neuromuscular control of CAI.

ABSTRACT

Wulingsan, a classic prescription first recorded in the Treatise on Cold Damage (Shang Han Lun) by ZHANG Zhong-jing for patients with water retention syndrome due to the disturbance of Qi transformation in bladder, has often been modified by ancient and modern doctors for the treatment of renal diseases. It produced satisfactory outcomes without inducing adverse reactions. The databases including China National Knowledge Infrastructure (CNKI), Wanfang Data Knowledge Service Platform, and PubMed were searched for articles concerning the clinical application and pharmacological mechanism of Wulingsan in the treatment of renal diseases published by domestic and foreign scholars in recent years. The summary of the included articles revealed that Wulingsan has been widely employed for the treatment of renal edema, diabetic nephropathy, acute and chronic glomerulonephritis, nephrotic syndrome, renal insufficiency, abnormal urination, etc., and the resulting outcomes were satisfactory. Wulingsan alone or in combination with western medicine yielded better clinical outcomes than the western medicine alone. In the exploration of its pharmacological mechanism, there existed some individual reports by Korean scholars, while Chinese scholars tended to work as a team. After years of research, each team has found that Wulingsan was superior to Wulingsan decoction in excreting water, which might be related to the absence of massive low-polarity and volatile components in the decoction that were responsible for regulating the expression of aquaporins (AQP) in kidney of saline-loaded rats. Wulingsan regulates the endocrine state of rats with kidney yang deficiency, inhibits the reabsorption of water, increases the activity of Na+-K+-ATPase, and reduces the expression of AQP 1, AQP 2, AQP 3, and AQP 4. Besides, it bi-directionally regulates the human water metabolism, which is achieved via the dual-directional regulation of purinergic P2X3 receptor expression on bladder detrusor. The efficacy of Wulingsan in treating chronic renal failure is attributed to its protection against the damaged renal tissue, structure and fanction. Hence, this paper summarized the research progress of Wulingsan in the treatment of renal diseases in recent years, aiming to provide a reference for the clinical and basic exploration of Wulingsan against renal diseases in the future.

ABSTRACT

Objective:To systematically evaluate the effects of elastic resistance exercise on patients with chronic obstructive pulmonary disease (COPD). Methods:Randomized clinical trials about elastic resistance exercise for chronic obstructive pulmonary disease were searched in the Cochrane Library, PubMed, Web of Science, CNKI, VIP, Wanfang Database and Chinese Biomedical Literature Database from inception to September, 2020. Finally, eleven literatures were included, including 9 in English and 2 in Chinese, with a total of 485 patients. The quality of the articles was evaluated using the Cochrane Library systematic review criteria and Physiotherapy Evidence Database Scale, and the data were analyzed with RevMan 5.2. The system review was registered on PROSPERO (CRD42020208659). Results:There was no significant difference in 6 Minute Walking Distance (6WMD) (MD = 1.19, 95%CI -7.02 to 9.39, P = 0.78), COPD Assessment Test (CAT) (MD = -0.43, 95%CI -2.42 to 1.57, P = 0.68) and the muscle strength (MD = 0.23, 95%CI -1.06 to 1.52, P = 0.73), with no high heterogeneity, between elastic resistance exercise group and the conventional resistance exercise group (such as weight training machine, weight training). There was no significant difference in 6MWD (MD = 18.30, 95%CI -8.92 to 45.52, P = 0.19) and CAT (MD = 0.59, 95%CI -3.78 to 2.60, P = 0.72) between the elastic resistance exercise group and the non-resistance exercise group, however, the heterogeneity between them was high. Conclusion:Elastic resistance exercise may be potentially alternative to conventional resistance training. However, the effects of elastic resistance exercise on exercise endurance, quality of life and lung function are still unclear.

ABSTRACT

Anterior cruciate ligament (ACL) deficiency is a common clinical injury that seriously affects the function of the knee. Although ACL reconstruction (ACLR) can rebuild the structure of the ACL, it does not restore the normal kinematics and dynamics of the knee. In recent years, motion analysis has been widely applied in sports science and rehabilitation. In clinics, motion analysis is mainly applied to the evaluation of clinical outcomes of the ACLR knee. It is also a scientific objective tool to assess the potential risk factors of secondary injury and early onset of osteoarthritis after ACLR. In-depth understanding of the knee functional kinematics and kinetics after ACLR will significantly improve the efficacy of current ACL deficiency treatment and rehabilitation regime. This paper reviewed studies of the knee joint motion analysis after ACLR in the past decade. The current knowledge of the ACLR knee motion from studies using optical motion capture and biplanar fluoroscopic imaging system based tracking technique was synthesized. Clinical studies have reported that regardless of the type of graft and surgical technique of the ACLR, the postoperative knee still exhibits abnormal kinematics in functional tasks, but the relationship between early-onset OA and the biomechanical changes in the ACLR knee remains unknown. Restoring the rotational stability of the knee is still one of the biggest challenges in clinical ACLR surgery. Future studies should explore the impact of ACLR on the biomechanical mechanisms of the knee joint through an appropriate research design and long-term follow-up, and help clinicians and physiotherapists to develop more effective interventions.

ABSTRACT

BACKGROUND: The proximal femur anti-rotation intramedullary nail is an ideal internal fixation method for the treatment of intertrochanteric fractures, but there is still a 6% to 21% failure rate of internal fixation. Tip-apex distance is considered as an important cause of postoperative proximal femur anti-rotation intramedullary nail failure. Tip-apex distance is the sum of the distance from the tip of lag screw to the vertex of femoral head measured on anteroposterior and lateral X-ray films. Most scholars now believe that the tip-apex distance of head pulp nail ≤ 25 mm has a good prognosis, but there is still a lot of controversy. OBJECTIVE: To investigate the biomechanical differences of the treatment of intertrochanteric femoral fractures by proximal femoral anti-rotation intramedullary nail with different tip-apex distances and provide a new idea and experimental basis for the clinical treatment of intertrochanteric fractures. METHODS: CT data of one volunteer were imported into Mimics 19.0 and Geomagic studio 2017 software to extract and optimize the three-dimensional model of the right femur. SolidWorks 2017 software was used to draw the internal fixation model and assemble it with the AO2.1 type fracture femur model in different tip-apex distance according to the standard operation technology. Totally four models with tip-apex distance of 15, 20, 25, and 30 mm were obtained and imported into HyperMesh 14.0 software to mesh. The four models were imported into Abaqus 2016 software in inp format to set up with material property parameters, boundary conditions and applied loads. Finally, operation results were viewed in the visualization module. RESULTS AND CONCLUSION: (1) When tip-apex distance was too large (30 mm) or too small (15 mm), the stress at the proximal femur was reduced, but the displacement of the femur head and neck fragment and the lesser trochanter fragment was larger and the inversion was more serious. When tip-apex distance was in the middle (20, 25 mm), the displacement and varus of femoral head and neck fragment were small, and the lesser trochanteric fragment was basically unshifted. (2) In the treatment of intertrochanteric femur fractures, tip-apex distance should be adjusted to 20-25 mm to reduce displacement and obtain a better biomechanical effect.

ABSTRACT

OBJECTIVE@#To explore the effect of pretreatment of neuroblastoma cells with hot water extract of Korean ginseng on MNNG-induced parthanatos and its mechanism.@*METHODS@#Neuroblastoma SH-SY5Y cells were pretreated with 1 mg/L hot water extract of Korean ginseng before induction with 250 μmol/L MNNG for 1 h or 4 h. CCK-8 and cell flow cytometry were used to detect cell survival rate. Western blotting was used to detect the changes in poly(ADP-ribose) (PAR) expression in the treated cells. Immunofluorescence assay was used to detect nuclear distribution of apoptosis-inducing factor (AIF), and flow cytometry was used to detect the level of reactive oxygen species (ROS) in the cells.@*RESULTS@#Compared with the blank control cells, MNNG-treated SH-SY5Y cells showed significantly decreased survival rate as the concentration of MNNG and the stimulation time increased ( < 0.05). Stimulation with MNNG also resulted in significantly increased expression of PAR protein in the cells ( < 0.05). Pretreatment of the cells with hot water extract of Korean ginseng obviously inhibited MNNG-induced cell death and significantly reduced AIF expression and nucleation in the cells ( < 0.05). MNNG stimulation significantly increased ROS level in the cells, which was decreased significantly by pretreatment of the cells with the extract ( < 0.05).@*CONCLUSIONS@#Pretreatment with hot water extract of Korean ginseng reduces MNNG-induced parthanatos and ROS production in SH-SY5Y cells.

ABSTRACT

Objective To evaluate the effects of water body environments on the microbial community of Oncomelania hupensis snails in marshlands of the eastern Dongting Lake where natural extinction of O. hupensis snails are found, so as to explore the correlation between the natural extinction of O. hupensis snails and the microbial community in snails. Methods Snails were caged water bodies in the Qianliang Lake marshland (Qianliang Lake regions) where natural extinction of snails was found and in the Junshan Park marshland (Junshan Park regions) in the eastern Dongting Lake for 30 days, and then all snails were collected and identified for survival or death. DNA sequencing of the fungi and bacteria was performed in snails before and after immersion in waters, and the biodiversity and abundance were analyzed. Results The survival rates of O. hupensis snails were 28.0% (70/250) and 64.8% (162/250) in Qianliang Lake regions and Junshan Park regions 30 days after immersion in waters, respectively (χ2 = 81.365, P < 0.01). The number of the fungal community and the biodiversity of the bacterial community were both greater in snails caged in Qianliang Lake regions post-immersion than pre-immersion, and there was a significant difference in the structure of the fungal and bacterial communities. The microbial community with a significant difference included Flavobacteriaceae, which was harmful to O. hupensis snails. Conclusion The water body environment affects the composition of the microbial community in O. hupensis snails in marshlands with natural snail distinction around the eastern Dongting Lake; however, further studies are required to investigate whether the natural distinction of snails is caused by water body environments-induced changes of the microbial spectrum in O. hupensis snails.

ABSTRACT

Objective To investigate the reasons of natural extinction of Oncomelania hupensis snails by comparing the difference of the water chemical properties between the snail-breeding marshland and non-snail marshland in Eastern Dongting Lake areas. Methods Three adjoining marshlands in Eastern Dongting Lake areas were selected, one was a snail-breeding marshland, the second was a non-snail marshland, and the third was a boundary zone between them. During the periods of water-rising season, wet season and water-falling season, the water samples from the bottom of 0.5 m were collected by the systematic random sampling method (200 m × 200 m grid method). In addition, the water samples below the surface of 0.5 m were also collected in the wet season. The physicochemical indexes of the sampling water were detected. Results According to the Surface Water Environmental Quality Standard (GB 3838–2002), the water quality indicators which were less than the detection limit were Cr, Cd, Zn, Hg, Cu, S- and CN- during the three sampling seasons. The pollutants which were slightly higher than Grade III but lower than Grade IV (Hg ≤ 1 μg/L, TP ≤ 0.1 mg/L) were Hg and TP. The pollutants below the Grade III included As and F-. The temperature and pH values were within the Grade III. The oxygen consumption indicators covering DO, COD and BOD in the water-rising season and wet season were within the Grade III, while the value of BOD was beyond the scope of Grade III but was between Grade Ⅳ (≤ 6 mg/L) and Grade V (≤ 10 mg/L) in the water-falling season. The differences of the water pollution indexes between the snail-breeding marshland and non-snail marshland were statistically significant, and these indexes included F-, As and pH in the water-rising season (P < 0.05); pH, BOD and Fat the surface layer in the wet season (P < 0.05); F- at the bottom in the wet season (P < 0.05); TP and F- in the water-falling season. Moreover, the above-mentioned indexes in the non-snail marshland of Qianliang Lake were higher than those in the snail-breeding marshland of Junshan Park. Thus, F- was the only index which had statistical differences in every sampling season, and the concentration of F- at the non-snail marshland was also higher than that at the snail-breeding marshland. In the water-rising season, the pH value at the non-snail marshland exceeded the suitable range for the growth of snails (6.8 to 7.8). Conclusion In the Eastern Dongting Lake area, the high concentration of F- and pH of water may be responsible for the natural extinction of O. hupensis snails.
